A successful proof of concept by the Kim lab (CTAR consortium), soon to appear in Nature Communications, marks another HIRN advance in exploiting CRISPR/Cas9 gene editing to answer important biological questions. Previously, several HIRN groups have contributed to improvements to the CRISPR/Cas9 technique and pioneered its usage for the engineering of beta cell lines and alpha cell lines. What the Kim lab recently demonstrated is a CRISPR/Cas9 strategy that can edit primary adult human islets, which should provide a great model system for later studies.
Their experiments, targeting both coding and non-coding regions, establish effectiveness and function as examples of how their lentiCRISPR-pseudoislet system can be applied. The first experiment removed PDX1 in primary beta cells within cultured pseudoislets without problematic side effects (off-target modifications). Given that PDX1 is a transcription factor important in beta cell function, this yields edited pseudoislets that could be assayed directly or transplanted to examine loss-of-function phenotypes. Their method to achieve loss-of-function was generalizable, as demonstrated by another experiment to remove a coding region of KCNJ11. The second set of experiments focused on non-coding variants that have been associated with diabetes risk but can be difficult to study functionally in cell lines. Using their lentiCRISPR technique, GWAS-linked variants in the KCNJ11-ABCC8 and the SIX2-SIX3 loci were modified to reveal regulatory interactions that other approaches had earlier failed to elucidate. Overall, the work by the Kim group shows how to wield CRISPR/Cas9 in a powerful system, and getting it to work on primary human islets should lead to excitement about what can be done next.

Don't miss this great opportunity to learn best practices to enhance rigor and reproducibility and learn the basics of good data management by following the FAIR (Findable, Accessible, Interoperable, Reusable) data principles! The program is a 6-week online program, and selected students will receive $1,000 awards. Research projects should be in the areas of the NIDDK including diabetes, obesity, nutrition, digestive, kidney, liver, urologic, endocrine, metabolic, bone, and blood diseases. Preference will be given to those projects that would most benefit from the types of services offered by dkNET, e.g., projects that involve antibodies, genetically modified organisms, and ‘omics’ data. Program Director for Diabetes Behavioral Science
The National Institute of Diabetes and Digestive and Kidney Diseases (NIDDK) is seeking a clinical behavioral scientist to serve as Program Director for Diabetes Behavioral Science. The National Institute of Diabetes and Digestive and Kidney Diseases is hiring program officials with expertise in translational research in any of the following four topic areas as they specifically relate to key questions in diabetes, endocrine or metabolic diseases:
· Integrative physiology of human diabetes, including physiology of counter-regulatory responses to hypoglycemia, mechanisms involved in regulation of peripheral metabolism, autonomic nervous system regulation of glucose and lipid metabolism, and basic and clinical studies investigating effects of bariatric surgery on diabetes and diabetes-related outcomes;

Faculty Position
The Diabetes Clinical Research Program (DCRP) at Benaroya Research Institute at Virginia Mason (BRI) seeks to recruit outstanding applicants for a new faculty position. The appointment will be at the at the assistant, associate or full member level, commensurate with experience, training and achievement. The DCRP is an international leader in type 1 (autoimmune) diabetes research. The Indiana Biosciences Research Institute (IBRI) Diabetes Center has a postdoctoral position available. We are looking for highly motivated researcher to join the IBRI in the lab of Dr. Decio Eizirik. This lab aims to clarify the mechanisms of pancreatic beta cell dysfunction and death in type 1 diabetes mellitus, with focus on endoplasmic reticulum stress, alternative splicing and other mechanisms that may contribute to beta cell death and generation of neoantigens. Postdoctoral Position
The Montreal Diabetes Research Center at the University of Montreal, has two Postdoctoral positions available at the Rutter laboratory. The laboratory studies pancreatic beta cell biology and its perturbations in type 2 diabetes (Rutter et alDiabetologia, 63(10):1990-1998. doi: 10.1007/s00125-020-05205-5, 2020). The objectives of the two projects are to explore the role and regulation of interactions between beta cells within the pancreatic islet using live cell imaging and optogenetics (Project 1), and to understand how genomic variants associated with type 2 diabetes impact pancreatic beta cell function (Project 2). The successful applicants will hold a PhD or equivalent. Experience in islet biology and/or glucose homeostasis, calcium signaling and image analysis (Project 1) or CRISPR/Cas9 genome editing, culture of human-derived beta cells or directed differentiation of embryonic stem cells towards a beta cell fate would be advantageous. A postdoctoral research associate position is available in the Hughes Lab at the Washington University School of Medicine to study primary cilia in islet cells. The laboratory combines standard molecular and cell biology with advanced imaging techniques to explore novel roles of cilia in mouse and human islets. We have generated new cilia knockout mouse models with strong diabetic phenotypes and are looking for someone to lead the investigation of cilia-mediated signaling mechanisms in islet hormone secretion. Postdoctoral Position
The Arrojo e Drigo lab focuses on the mechanisms regulating post‐mitotic cell homeostasis and longevity. Many postmitotic cells can be as old as the organism, which makes them remarkably long‐lived. We study long‐lived cells in different organs and explore their developmental trajectory, structure‐function and aging patterns. Our lab uses a combination of single cell sequencing and high‐resolution light, electron and isotope microscopy platforms to overlay cell structure‐function with molecular identity, age and metabolism.
