MacConkey MUG Agar is a moderately selective medium used for the isolation of Gram-negative enteric bacilli from feces, urine, food, wastewater and other materials of sanitary importance for the differentiation of lactose fermenting organisms from non fermenting ones.
The medium, which contains 4-methylumbelliferyl-beta-D-glucuronide (MUG), allows to presumptively identify Escherichia coli from the primary plating medium within five minutes by observing for fluorescent colonies under long-wave UV light.
Organisms which do not ferment lactose, such as Salmonella, Shigella and Proteus spp, form colourless or clear colonies. Lactose fermenters, such as E. coli and Klebsiella spp, develop pink to red colonies with or without an halo of bile precipitation.
Beta-glucuronidase positive colonies fluoresce blue-green under UV light. Negative colonies do not show any fluorescence. Enterococci, Staphylococci and other Gram-positive bacteria are partially to completely inhibited.
