Protein Conformational Array (PCA) technology was developed based on the principle of epitope mapping in immunology. It offers high sensitivity, systematic coverage and high throughput for mAb Higher Order Structure (HOS) analysis. It can detect structural changes induced by physical and chemical stress such as light, temperature, pH, glycosylation, oxidation and aggregation. Using the NIST (National Institute of Standards and Technology) mAb as an example and tested with 5 different formulation conditions, it was demonstrated that PCA can detect mAb HOS changes at molecular level, providing valuable information for the selection of best stabilizing formulation for a specific mAb.
