FAQs concerning ELISA Substrates

Q: 
What detection range can I expect when using an HRP colorimetric substrate?
A:
With ABTS, you can expect linear results between 10 µg/mL and 1 ng/mL.  With TMB, you can expect linear results between 1 µg/mL and 0.1 ng/mL.  Detection ranges are assay-dependent and assays must be optimized based on the amount of target being detected. Q:
Which enzyme offers the best ELISA sensitivity: HRP or AP?
A:
HRP substrates provide much better sensitivity (low nanogram/high pictogram amounts) than AP substrates (nanogram amounts). HRP conjugates are the best choice when high sensitivity is required.
Q:
How do I stop my colorimetric substrate reaction?
A:
Substrate reactions can be stopped using Stop Solutions which are added directly to the microwells. Stop solutions work by changing the pH, chelating the enzyme, or through denaturation. SeraCare offers at least one stop solution for all our colorimetric substrates. Learn more.
