US FDA approves Sanofi and Regeneron’s

Dupixent to treat prurigo nodularis

US Food and Drug Administration (FDA) has approved Sanofi and Regeneron’s Dupixent(dupilumab), a fully human monoclonal antibody can block the interleukin-4 (IL-4) and interleukin-13 (IL-13) pathways signalling, to treat prurigo nodularis, a chronic inflammatory skin disease.

The regulatory approval is based on the data obtained from two Phase III trials, PRIME and PRIME2, which evaluated the safety and efficacy of Dupixent in adults with prurigo nodularis.

In order to accelerating the drug developments of IL-4/ IL-4Rα, ACROBiosystems has developed IL-4Rα: IL-4 inhibitor screening kit using sandwich ELISA method based on unique biotinylated IL-4 and IL-4Rα proteins, which can used in antibody/small molecular inhibitor screening and QC release.

Features

★ Accurate and reliable: Verified raw materials of the kit with high batch-to-batch consistency ensures stable and reliable experimental results, which is more conducive to long-term development and optimization of verification experiments.

★ Fast and efficient: By employing the biotin pre-labeled proteins, we eliminate the time-consuming labeling process, and greatly simplify the experimental procedures. Besides, saving time for method establishment and validation as the method is fully validated.

★ Reduce interference: The matrix interference is small, and the interference caused by DMSO to the system is verified to ensure that kit can be used to screen small-molecules/macromolecular inhibitors.

★ High repeatability: Our biotin labeling platform produces biotinylated proteins are using site-specific labeling, and the more stable labeling method ensures minimal lot-to-lot variation and the stability of the kit.

Typical Data

Screening IL-4Rα inhibitors that bind to human IL-4 with ELISA kit (Cat. No. EP-132)

Serial dilutions of Human IL-4(Cat. No.EP132-C03) (1:1 serial dilution, from 5 μg/mL to 0.01 μg/mL (295.86-0.58 nM)) was added into IL-4 R alpha: IL-4-Biotin binding reactions. The assay was performed according to the above-described protocol. Background was subtracted from data points prior to log transformation and curve fitting (QC tested).
